Make.conf é o arquivo de configuração principal do Portage, por conter muitas variáveis. Por exemplo, o melhor conhecimento: CFLAGS, CHOST, CXXFLAGS, USE, INPUT_DEVICES, VIDEO_CARDS, LINGUAS, GENTOO_MIRRORS. Há também muitas variáveis opcionais, como por exemplo: PORTDIR_OVERLAY. Nesta distribuição Linux, o usuário compila o código fonte localmente de acordo com a sua configuração escolhida, a fim de melhorar o seu sistema e também para aprender mais sobre o Linux.
